Norgen’s Urine Collection and Preservation Tube contains a dried storage buffer designed for the rapid preservation of nucleic acids and proteins from fresh urine specimens. The buffer prevents the growth of Gram-negative and Gram-positive bacteria and fungi, and also inactivates viruses allowing the resulting non-infectious samples to be handled and shipped safely. In addition, the buffer eliminates the need to immediately process or freeze samples and allows the samples to be shipped to centralized testing facilities at ambient temperature. The components of the buffer allow samples to be stored for one year under conditions where RNA/DNA/protein degradation would occur normally. Norgen’s Urine Collection and Preservation Tube is intended to be used in clinical laboratories with the ability to preserve samples for use in downstream diagnostic assays.

To collect samples, the patient is advised to collect a maximum of 50 mL from the midstream urine sample directly in Norgen’s Urine Collection and Preservation Tube (collecting the first flow of urine is not recommended). The Urine sample should be mixed gently by inversion with the coloured dried storage buffer. Norgen’s Urine Collection and Preservation Tube is recapped tightly and labelled with the patient’s ID, collection date and hospital/clinic.

APPLICATIONS

The preservation buffer is compatible with most DNA, RNA and protein isolation methods. Samples stored in the Sample Collection and Preservation Tube has been used successfully with all of Norgen Biotek’s DNA, RNA and Protein isolation kits and reagents. Urinary proteins obtained from samples stored in the buffer will be suitable for applications such as Western blotting or 2D gel electrophoresis as well as applications which require native protein.

Two 50 mL urine samples were collected using Norgen’s Urine Collection and Preservation Tubes, and one tube was stored at 4°C and one tube was stored at room temperature. Aliquots of urine (2 mL) were removed after 2, 4, 6, 8, 10 ,12, 14 and 16 weeks, and the urinary proteins were isolated. The purified proteins were then run on 12% SDS-PAGE gels for visual analysis. Panel A contains the proteins isolated from urine stored at 4°C, while Panel B contains the proteins isolated from urine stored at room temperature. In both cases, Lane A corresponds to 2 weeks, Lane B corresponds to 4 weeks, Lane C corresponds to 6 weeks, Lane D corresponds to 8 weeks, Lane E corresponds to 10 weeks, Lane F corresponds to 12 weeks, Lane G corresponds to 14 weeks and Lane H corresponds to 16 weeks. As it can be seen, total urinary proteins isolated from samples stored at either 4°C or at room temperature in Norgen’s Urine Collection and Preservation Tubes were of a very good quality with no signs of protein degradation.

The purified DNA was then used as the template in a PCR reaction for the amplification of the GAPDH house keeping gene. The PCR products were then run on 1.7% agarose gels for visual analysis. Panel A contains the PCR resulst from when the DNA was isolated from urine stored at 4°C, while Panel B contains the PCR results when DNA was isolated from urine stored at room temperature. In both cases, Lane A corresponds to 2 weeks, Lane B corresponds to 4 weeks, Lane C corresponds to 6 weeks, Lane D corresponds to 8 weeks, Lane E corresponds to 10 weeks, Lane F corresponds to 12 weeks, Lane G corresponds to 14 weeks and Lane H corresponds to 16 weeks. As it can be seen the GAPDH housekeeping gene was successfully amplified from DNA isolated from 2 mL of urine that was stored at either 4°C or at room temperature, indicating the high quality of the purified DNA.
