Soil DNA Isolation Kit

Norgen’s Soil DNA Isolation Kit provides PCR-inhibitor free DNA through a convenient and rapid method for the detection of microorganisms from soil samples. All common types of soil samples can be processed with this kit, including common soil samples such as clay, loam and sandy soil.  For difficult soil samples with high humic acid content such as compost and manure, we recommend the use of Norgen's Soil DNA Isolation Kit (High Humic Acid) - Cat# 49300. Norgen's Soil DNA Isolation Kit removes all traces of humic acid using the provided Bead Tubes and a combination of chemical and physical homogenization and lysis. A simple and rapid spin column procedure is then used to further purify the DNA.

Purification is based on spin column chromatography using Norgen's resin as the separatiom matrix.  Briefly, the soil sample is lysed using Lysis Solution in combination with the provided Bead Tubes.  The released DNA is then bound to Norgen's column in the presence of Binding Solution (BIND).  Under these conditions only the DNA will bind to Norgen's resin while most of the contaminating RNA and cellular proteinaceous components are removed in the flowthrough.  The bound DNA is then washed to remove any remaining impurities (WASH).  Lastly, the purified inhibiitor-free DNA is eluted into 50 - 100 µL of the provided Elution Buffer or water (ELUTE).  Please see procedure flowchart to the right.  

Total genomic DNA can be isolated and purified from all the various microorganisms found in soil, such as bacteria, fungi and algae. The purified DNA is of the highest quality and is fully compatible with downstream PCR applications, as all humic acid substances and PCR inhibitors are removed during the isolation.
