Collection Protocol

CAUTION: sharp needle is located under the cap warning label.
DO NOT remove the warning label on the top of the cap.
DO NOT open the vacutainer tubes at any time. They are pressurized and contain stabilization reagent.

1. 

   Open the Cup without removing any sticker: unscrew the yellow cap and place it on a clean, flat surface with the straw facing up. DO NOT touch the inside of the cap or straw.

2. 

   Collect the urine sample making sure not to go above the 60mL mark, then close the cup.

3. Remove the top sticker on the cup. Push the vacutainer tube onto the cup needle and hold firmly until filling stops. Repeat this with the second tube.

   Tube does not fill / no flow --> improper seating, cap not fully pierced, or misalignment.

4. After each vacutainer is filled with urine, mix by inverting the vacutainer tube 3 times.

5. 

   Sample tubes can be submitted. Left over urine can be poured into the toilet. Yellow cap should be discarded carefully in a puncture proof container and the cup can be discarded in household trash.

Please Note: The vacutainer tubes each have 10 mL maximum capacity. However, due to the vacuum system, expect each tube to fill with approximately 9 mL of urine.

Figure 1. Stability of Viral DNA in Urine sample preserved with Urine collection and preservation system.Urine sample was spiked with Adenovirus and added to Urine collection cups. Urine sample was then transferred to vacutainer tubes containing the preservative using the integrated transfer device. Tubes containing the sample were stored at 55℃ for the mentioned time points. Norgen’s Urine DNA isolation kit 48800 was used to isolate DNA and qPCR was performed to detect Adenovirus DNA. Viral DNA was stable for 92 days at 55℃ (equivalent to 2 years at room temperature).

Urine sample was then transferred to vacutainer tubes containing the preservative using the integrated transfer device. Tubes containing the sample were stored at 55℃ for the mentioned time points. Norgen’s Urine DNA isolation kit 48800 was used to isolate DNA and 16S targeting qPCR was performed to detect bacterial DNA. Bacterial DNA was stable for 92 days at 55℃ (equivalent to 2 years at room temperature).

Urine sample was spiked with S. cerevisiae at 2 x 10⁴ CFU/mL and added to Urine collection cups. Urine sample was then transferred to vacutainer tubes containing the preservative using the integrated transfer device. Tubes containing the sample were stored at 55℃ for the mentioned time points. Norgen’s Urine DNA isolation kit 48800 was used to isolate DNA and 18S targeting qPCR was performed to detect fungal DNA. Fungal DNA was stable for 92 days at 55℃ (equivalent to 2 years at room temperature).

Norgen’s Urine RNA isolation kit 29600 was used to isolate RNA and Rnase P targeting qPCR was performed to detect RNA. RNA was stable for 92 days at 55℃ (equivalent to 2 years at room temperature).
