Supporting Data
Figure 1. Norgen’s Plasma/Serum cfc-DNA Advanced Purification Kit was used to purify circulating DNA from 4 mL plasma from blood collected on EDTA as an anticoagulant from two different donors. Panel A and B shows the Bioanalyzer High Sensitivity DNA Chip trace for the cfc-DNA purified using Norgen’s kit as compared to Panel C and D representing the cfc-DNA purified by Competitor Q's kit which is a Magnetic Bead based kit. Norgen's kit showed the highest amount of the correct cfc-DNA size (Mono-nucleosomal cfc-DNA) with minimal high molecular weight gDNA contamination as opposed to the cfc-DNA profile obtained from Competitor Q's kit which showed low amount of the correct cfc-DNA with a bias toward binding higher molecular weight gDNA sizes.
Figure 2. Norgen’s Plasma/Serum cfc-DNA Advanced Purification Kit was used to purify circulating DNA from 4 mL plasma from blood collected on EDTA as an anticoagulant from three different donors. The concentration of the Mono-nucleosomal cf-DNA (representing the correct cfc-DNA size) was measured using the Agilent Bioanalyzer High Sensitivity DNA Chip. Norgen's kit showed the highest Mono-nucleosomal cfc-DNA concentration with an average of 182.7 pg/µL ±13 as compared to the average concentration obtained from Competitor Q's kit which was 31.5 pg/µL ±3.1.
Figure 3. Norgen’s Plasma/Serum cfc-DNA Advanced Purification Kit was used to purify circulating DNA from 1mL, 2mL, 4 mL and 6mL plasma prepared from blood collected on EDTA as an anticoagulant. 2 microlitres of the purified DNA was then used as the template in qPCR reactions to assess the linearity of the purified cfc-DNA by targeting a short ALU gene target (115 bp) representing the mono-nucleosomal cfc-DNA population. Norgen's Plasma/Serum cfc-DNA Advanced Purification Kit showed an excellent linearity between all plasma volumes with a % of linearity of more than 90% indicating efficient cfc-DNA recovery from all plasma volumes used.
